Probably depends on how long the 2 min lasts, and if there's an appreciable initial damage component.
Ideally you likely want to have your trinket affect as much of your big CD as it can, so if your CD lasts for 30 seconds and your trinket lasts for 20 it'd be best to pop it afterward.

Create a scheme of button presses in your mind that relate to a specific kinda action. For example ill use monk

1 = Generator (tpalm)
2 = Spammable Spender (blackout kick)
3/4 = Higher value spender (RSK/FoF)
5 = WDP
Shift+1 = major cooldown 1 (SEF/Tiger)
Shift +2 = major cooldown 2 (SEF/Tiger)
.....
Q = aoe spender (SPK)
E = Interrupt

Once you associate the button press with an action you will start building muscle memory. Also once you remember basic buttons hiding them is a good idea to force attention to the things you dont remember. Dont sweat too much about being perfect though preach played this game for 20 years and still cant parse above greys
